Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_144997.7(FLCN):c.302A>G (p.Glu101Gly), citing Ambry Variant Classification Scheme 2023: The p.E101G variant (also known as c.302A>G), located in coding exon 2 of the FLCN gene, results from an A to G substitution at nucleotide position 302. The glutamic acid at codon 101 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
