This variant lies in the DNM1 gene (transcript NM_004408.4) at coding-DNA position 1891, where G is replaced by C; at the protein level this means replaces glycine at residue 631 with arginine — a missense variant. Submitter rationale: Variant summary: DNM1 c.1891G>C (p.Gly631Arg) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. 4/4 computational tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 6.6e-06 in 150974 control chromosomes (gnomAD v3.1.2). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance.
