Uncertain significance for Autoimmune lymphoproliferative syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000043.6(FAS):c.161A>G (p.His54Arg), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt FAS protein function. This variant has not been reported in the literature in individuals with FAS-related conditions. This variant is present in population databases (rs759152311, ExAC 0.006%). This sequence change replaces histidine with arginine at codon 54 of the FAS protein (p.His54Arg). The histidine residue is weakly conserved and there is a small physicochemical difference between histidine and arginine.
