NM_005506.4(SCARB2):c.1208C>T (p.Thr403Ile) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T403I variant (also known as c.1208C>T), located in coding exon 10 of the SCARB2 gene, results from a C to T substitution at nucleotide position 1208. The threonine at codon 403 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
