NM_002691.4(POLD1):c.3225C>G (p.Asp1075Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1075E variant (also known as c.3225C>G), located in coding exon 26 of the POLD1 gene, results from a C to G substitution at nucleotide position 3225. The aspartic acid at codon 1075 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
