Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.3434G>A (p.Arg1145Lys), citing Ambry Variant Classification Scheme 2023: The p.R1145K variant (also known as c.3434G>A), located in coding exon 5 of the MSH6 gene, results from a G to A substitution at nucleotide position 3434. The arginine at codon 1145 is replaced by lysine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
