Pathogenic for Charcot-Marie-Tooth disease type 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001370298.3(FGD4):c.1051del (p.Leu351fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in FGD4 are known to be pathogenic (PMID: 17564972). This variant has not been reported in the literature in individuals with FGD4-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Leu214Phefs*15) in the FGD4 gene. It is expected to result in an absent or disrupted protein product.
