NM_004064.5(CDKN1B):c.397C>T (p.Pro133Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P133S variant (also known as c.397C>T), located in coding exon 1 of the CDKN1B gene, results from a C to T substitution at nucleotide position 397. The proline at codon 133 is replaced by serine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
