NM_000070.3(CAPN3):c.791G>A (p.Cys264Tyr) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the CAPN3 gene (transcript NM_000070.3) at coding-DNA position 791, where G is replaced by A; at the protein level this means replaces cysteine at residue 264 with tyrosine — a missense variant. Submitter rationale: Variant summary: CAPN3 c.791G>A (p.Cys264Tyr) results in a non-conservative amino acid change located in the Peptidase C2, calpain, catalytic domain (IPR001300) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251186 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.791G>A has been reported in the literature in the heterozygous state in an individual affected with Autosomal Recessive Limb-Girdle Muscular Dystrophy, without strong evidence for causality (Krahn_2006). This report does not provide unequivocal conclusions about association of the variant with Limb-Girdle Muscular Dystrophy. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 16650086). ClinVar contains an entry for this variant (Variation ID: 856576). Based on the evidence outlined above, the variant was classified as uncertain significance.
