Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_032638.5(GATA2):c.224C>T (p.Ala75Val), citing Ambry Variant Classification Scheme 2023: The p.A75V variant (also known as c.224C>T), located in coding exon 1 of the GATA2 gene, results from a C to T substitution at nucleotide position 224. The alanine at codon 75 is replaced by valine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
