Likely pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005802.5(TOPORS):c.2569_2570del (p.Arg857fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Arg857Glufs*15) in the TOPORS gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 189 amino acid(s) of the TOPORS protein. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with TOPORS-related conditions. ClinVar contains an entry for this variant (Variation ID: 856353). This variant is located in a region of the TOPORS protein where a significant number of TOPORS nonsense and frameshift mutations have been reported in association with autosomal dominant retinitis pigmentosa (PMID: 35579903, 17924349, 32531858). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
