Uncertain significance for Hereditary spastic paraplegia 45 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001351169.2(NT5C2):c.1259A>G (p.Gln420Arg), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with NT5C2-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glutamine with arginine at codon 420 of the NT5C2 protein (p.Gln420Arg). The glutamine residue is highly conserved and there is a small physicochemical difference between glutamine and arginine.
