NM_001042492.3(NF1):c.5493G>A (p.Trp1831Ter) was classified as Pathogenic for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W1810* pathogenic mutation (also known as c.5430G>A), located in coding exon 37 of the NF1 gene, results from a G to A substitution at nucleotide position 5430. This changes the amino acid from a tryptophan to a stop codon within coding exon 37. This variant has been reported in multiple individual(s) in neurofibromatosis type 1 cohorts, but clinical details were limited (Wang HF et al. J Clin Invest, 2011 Dec;121:4820-37; Sabbagh A et al. Hum Mutat, 2013 Nov;34:1510-8). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
