Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.748A>G (p.Lys250Glu), citing Ambry Variant Classification Scheme 2023: The p.K250E variant (also known as c.748A>G), located in coding exon 7 of the TSC2 gene, results from an A to G substitution at nucleotide position 748. The lysine at codon 250 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
