This variant lies in the MUSK gene (transcript NM_005592.4) at coding-DNA position 1099, where C is replaced by G; at the protein level this means replaces arginine at residue 367 with glycine — a missense variant. Submitter rationale: This sequence change replaces arginine with glycine at codon 367 of the MUSK protein (p.Arg367Gly). The arginine residue is highly conserved and there is a moderate physicochemical difference between arginine and glycine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with MUSK-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
