This variant lies in the SCN1A gene (transcript NM_001165963.4) at coding-DNA position 974, where A is replaced by G; at the protein level this means replaces tyrosine at residue 325 with cysteine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has been observed to be de novo in several individuals affected with clinical features consistent with an SCN1A-related condition (PMID: 23821540, Invitae).
