Pathogenic for Joubert syndrome 23; Short-rib thoracic dysplasia 14 with polydactyly — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001329943.3(KIAA0586):c.3668C>G (p.Ser1223Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Ser1276*) in the KIAA0586 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in KIAA0586 are known to be pathogenic (PMID: 26096313, 26166481, 26386044). This variant is present in population databases (rs776003330, gnomAD 0.01%). This premature translational stop signal has been observed in individual(s) with Joubert syndrome (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 854917). For these reasons, this variant has been classified as Pathogenic.
