Uncertain significance for Hereditary pheochromocytoma and paraganglioma — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_017841.4(SDHAF2):c.101T>C (p.Phe34Ser), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SDHAF2 gene (transcript NM_017841.4) at coding-DNA position 101, where T is replaced by C; at the protein level this means replaces phenylalanine at residue 34 with serine — a missense variant. Submitter rationale: This sequence change replaces phenylalanine with serine at codon 34 of the SDHAF2 protein (p.Phe34Ser). The phenylalanine residue is weakly conserved and there is a large physicochemical difference between phenylalanine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with SDHAF2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The serine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
