Pathogenic for Dilated cardiomyopathy 1HH; Myofibrillar myopathy 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004281.4(BAG3):c.974del (p.Pro325fs), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with BAG3-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change results in a premature translational stop signal in the BAG3 gene (p.Pro325Glnfs*17). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 251 amino acids of the BAG3 protein. This variant disrupts the C-terminus of the BAG3 protein. Other variant(s) that disrupt this region (p.Glu414*, p.Tyr441*) have been determined to be pathogenic (PMID: 25008357, Invitae). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. For these reasons, this variant has been classified as Pathogenic.
