This variant lies in the DOK7 gene (transcript NM_173660.5) at the canonical splice acceptor site of the intron immediately before coding-DNA position 773, where A is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects an acceptor splice site in intron 6 of the DOK7 gene. While this variant is not anticipated to result in nonsense mediated decay, it likely alters RNA splicing and results in a disrupted protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individuals with congenital myasthenic syndrome (PMID: 20012313, 36099689; internal data).
