This variant lies in the MYH11 gene (transcript NM_002474.3) at coding-DNA position 62, where T is replaced by C; at the protein level this means replaces isoleucine at residue 21 with threonine — a missense variant. Submitter rationale: Variant summary: MYH11 c.62T>C (p.Ile21Thr) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 2.2e-05 in 1613922 control chromosomes (gnomAD). The observed variant frequency is approximately 18 fold of the estimated maximal expected allele frequency for a pathogenic variant in MYH11 causing Aortopathy phenotype (1.3e-06), strongly suggesting that the variant is benign. To our knowledge, no occurrence of c.62T>C in individuals affected with Aortopathy and no experimental evidence demonstrating its impact on protein function have been reported.
