Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005228.5(EGFR):c.315G>C (p.Gln105His), citing Ambry Variant Classification Scheme 2023: The p.Q105H variant (also known as c.315G>C), located in coding exon 3 of the EGFR gene, results from a G to C substitution at nucleotide position 315. The glutamine at codon 105 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
