Pathogenic for Osteogenesis imperfecta type I — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000088.4(COL1A1):c.4325_4347del (p.Val1442fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change results in a frameshift in the COL1A1 gene (p.Val1442Alafs*101). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 23 amino acids of the COL1A1 protein and extend the protein by an additional 77 amino acids. For these reasons, this variant has been classified as Pathogenic. This variant results in an extension of the COL1A1 protein. Other variant(s) that result in a similarly extended protein product (p.Leu1445Profs*107) have been determined to be pathogenic (Invitae). This suggests that these extensions are likely to be causative of disease. This variant has not been reported in the literature in individuals with COL1A1-related conditions. This variant is not present in population databases (ExAC no frequency).
