This variant lies in the GALT gene (transcript NM_000155.4) at coding-DNA position 691, where C is replaced by G; at the protein level this means replaces arginine at residue 231 with glycine — a missense variant. Submitter rationale: In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. This variant disrupts the p.Arg231 amino acid residue in GALT. Another variant that disrupts this residue have been observed in individuals with GALT-related conditions (PMID: 14518827, 22944367, Invitae), suggesting that it is a clinically significant residue. As a result, variants that disrupt this residue are likely to be causative of disease. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies.
