Pathogenic for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001114753.3(ENG):c.626T>A (p.Leu209Ter), citing Ambry Variant Classification Scheme 2023. This variant lies in the ENG gene (transcript NM_001114753.3) at coding-DNA position 626, where T is replaced by A; at the protein level this means converts the codon for leucine at residue 209 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The p.L209* pathogenic mutation (also known as c.626T>A), located in coding exon 5 of the ENG gene, results from a T to A substitution at nucleotide position 626. This changes the amino acid from a leucine to a stop codon within coding exon 5. This mutation was reported in an individual with daily epistaxis and pulmonary arteriovenous malformations (McDonald J et al. Clin. Genet., 2011 Apr;79:335-44). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
