Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_005097.4(LGI1):c.1391C>T (p.Ser464Leu), citing Ambry Variant Classification Scheme 2023: The p.S464L variant (also known as c.1391C>T), located in coding exon 8 of the LGI1 gene, results from a C to T substitution at nucleotide position 1391. The serine at codon 464 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
