Pathogenic for Pigmentary pallidal degeneration — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001386393.1(PANK2):c.1352_1371del (p.Gly451fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the protein in which other variant(s) (p.Leu563Val and p.Leu566Val) have been observed in individuals with PANK2-related conditions (PMID: 28881514, 30681573). This suggests that this may be a clinically significant region of the PANK2 protein. This variant has been observed in individual(s) with clinical features of pantothenate kinase-associated neurodegeneration (Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 852118). This variant is not present in population databases (ExAC no frequency). This sequence change results in a frameshift in the PANK2 gene (p.Gly561Glufs*26). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 10 amino acids of the PANK2 protein and extend the protein by an additional 16 amino acids.
