NM_002691.4(POLD1):c.2475C>G (p.Asp825Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D825E variant (also known as c.2475C>G), located in coding exon 19 of the POLD1 gene, results from a C to G substitution at nucleotide position 2475. The aspartic acid at codon 825 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
