Submitter rationale: The p.T54M variant (also known as c.161C>T), located in coding exon 3 of the CTRC gene, results from a C to T substitution at nucleotide position 161. The threonine at codon 54 is replaced by methionine, an amino acid with similar properties. This alteration has been reported in a subject with pancreatitis whose mother was also noted to have pancreatitis. Funcational analysis of this alteration suggests an impact on protein secretion on endoplasmic reticulum stress (Tamura K et al. Proc. Natl. Acad. Sci. U.S.A., 2018 05;115:4767-4772). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
