Uncertain significance for Dyskeratosis congenita — the classification assigned by Ambry Genetics to NM_198253.3(TERT):c.120G>C (p.Gln40His), citing Ambry Variant Classification Scheme 2023: The p.Q40H variant (also known as c.120G>C), located in coding exon 1 of the TERT gene, results from a G to C substitution at nucleotide position 120. The glutamine at codon 40 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
