This variant lies in the SMAD4 gene (transcript NM_005359.6) at the canonical splice donor site of the intron immediately after coding-DNA position 787, where T is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: SMAD4 c.787+2T>C is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: Three predict the variant abolishes a 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 277066 control chromosomes (gnomAD).
