This variant lies in the MYBPC3 gene (transcript NM_000256.3) at coding-DNA position 3408, where C is replaced by G; at the protein level this means converts the codon for tyrosine at residue 1136 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. A different variant (c.3408C>A) giving rise to the same protein effect has been determined to be pathogenic (PMID: 25262865, 27532257). This suggests that this variant is also likely to be causative of disease. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Tyr1136*) in the MYBPC3 gene. It is expected to result in an absent or disrupted protein product.
