Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.131A>G (p.Glu44Gly), citing Ambry Variant Classification Scheme 2023: The p.E44G variant (also known as c.131A>G), located in coding exon 2 of the PMS2 gene, results from an A to G substitution at nucleotide position 131. The glutamic acid at codon 44 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
