Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_024675.4(PALB2):c.91A>G (p.Thr31Ala), citing Ambry Variant Classification Scheme 2023: The p.T31A variant (also known as c.91A>G), located in coding exon 2 of the PALB2 gene, results from an A to G substitution at nucleotide position 91. The threonine at codon 31 is replaced by alanine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
