NM_004304.5(ALK):c.4045G>C (p.Asp1349His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1349H variant (also known as c.4045G>C), located in coding exon 27 of the ALK gene, results from a G to C substitution at nucleotide position 4045. The aspartic acid at codon 1349 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
