This variant lies in the FOXP3 gene (transcript NM_014009.4) at coding-DNA position 1044, where G is replaced by T; at the protein level this means replaces tryptophan at residue 348 with cysteine — a missense variant. Submitter rationale: This sequence change replaces tryptophan with cysteine at codon 348 of the FOXP3 protein (p.Trp348Cys). The tryptophan residue is highly conserved and there is a large physicochemical difference between tryptophan and cysteine. This variant also falls at the last nucleotide of exon 10 of the FOXP3 coding sequence, which is part of the consensus splice site for this exon. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with FOXP3-related conditions.
