Uncertain significance for Lysinuric protein intolerance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003982.4(SLC7A7):c.860T>G (p.Met287Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine with arginine at codon 287 of the SLC7A7 protein (p.Met287Arg). The methionine residue is weakly conserved and there is a moderate physicochemical difference between methionine and arginine. This variant is present in population databases (rs755142027, ExAC 0.009%). This variant has not been reported in the literature in individuals affected with SLC7A7-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
