Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_012418.4(FSCN2):c.606G>C (p.Trp202Cys), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This sequence change replaces tryptophan with cysteine at codon 202 of the FSCN2 protein (p.Trp202Cys). The tryptophan residue is moderately conserved and there is a large physicochemical difference between tryptophan and cysteine. This variant has not been reported in the literature in individuals with FSCN2-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
