NM_000314.8(PTEN):c.54G>T (p.Glu18Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E18D variant (also known as c.54G>T), located in coding exon 1 of the PTEN gene, results from a G to T substitution at nucleotide position 54. The glutamic acid at codon 18 is replaced by aspartic acid, an amino acid with highly similar properties. This variant demonstrated wild type-like intracellular protein abundance in a massively parallel functional assay (Matreyek KA et al. Nat Genet, 2018 Jun;50:874-882). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
