This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 208, where G is replaced by A; at the protein level this means replaces glycine at residue 70 with arginine — a missense variant. Submitter rationale: The p.G70R variant (also known as c.208G>A), located in coding exon 2 of the FBN1 gene, results from a G to A substitution at nucleotide position 208. The glycine at codon 70 is replaced by arginine, an amino acid with dissimilar properties. A differnet variant affecting this codon (p.G70V, c.209G>T) has been detected in an individual with aortic dissection from a non-syndromic thoracic aortic aneurysm and dissection cohort (Guo J et al. Sci Rep, 2015 Aug;5:13115). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
