Uncertain significance for Generalized epilepsy with febrile seizures plus, type 9 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_052874.5(STX1B):c.833T>C (p.Leu278Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with serine at codon 278 of the STX1B protein (p.Leu278Ser). The leucine residue is highly conserved and there is a large physicochemical difference between leucine and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with STX1B-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C15"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
