Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.7360C>A (p.Pro2454Thr), citing Ambry Variant Classification Scheme 2023: The p.P2454T variant (also known as c.7360C>A), located in coding exon 59 of the FBN1 gene, results from a C to A substitution at nucleotide position 7360. The proline at codon 2454 is replaced by threonine, an amino acid with highly similar properties. This variant was reported in individual(s) with features consistent with aortic dilation (Ambry internal data). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
