Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001370259.2(MEN1):c.940C>T (p.Arg314Trp), citing Ambry Variant Classification Scheme 2023: The p.R314W variant (also known as c.940C>T), located in coding exon 6 of the MEN1 gene, results from a C to T substitution at nucleotide position 940. The arginine at codon 314 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
