Uncertain significance for Cardiomyopathy — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000257.4(MYH7):c.1956+1G>A, citing ACMG Guidelines, 2015. This variant lies in the MYH7 gene (transcript NM_000257.4) at the canonical splice donor site of the intron immediately after coding-DNA position 1956, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This variant causes a G to A nucleotide substitution at the +1 position of intron 17 of the MYH7 gene. Splice site prediction tools predict that this variant may have a significant impact on RNA splicing. Although this prediction has not been confirmed in published RNA studies, this variant is expected to result in an absent or disrupted protein product. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with MYH7-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Clinical relevance of loss-of-function MYH7 splice and truncation variants in autosomal dominant cardiovascular disorders is not clearly established. The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
