This variant lies in the WT1 gene (transcript NM_024426.6) at coding-DNA position 413, where C is replaced by G; at the protein level this means replaces proline at residue 138 with arginine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This sequence change replaces proline with arginine at codon 133 of the WT1 protein (p.Pro133Arg). The proline residue is weakly conserved and there is a moderate physicochemical difference between proline and arginine. This variant is not present in population databases (ExAC no frequency).
