This variant lies in the COL1A1 gene (transcript NM_000088.4) at the canonical splice donor site of the intron immediately after coding-DNA position 750, where T is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Donor and acceptor splice site variants typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in COL1A1 are known to be pathogenic (PMID: 7942841, 9295084, 9443882). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site, but this prediction has not been confirmed by published transcriptional studies. Disruption of this splice has been observed in an individual affected with osteogenesis imperfecta, type 1 (PMID: 28326186). This variant is not present in population databases (ExAC no frequency).
