This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 6471, where G is replaced by C; at the protein level this means replaces glutamic acid at residue 2157 with aspartic acid — a missense variant. Submitter rationale: The p.E2157D variant (also known as c.6471G>C), located in coding exon 44 of the ATM gene, results from a G to C substitution at nucleotide position 6471. The glutamic acid at codon 2157 is replaced by aspartic acid, an amino acid with highly similar properties. In an assay testing ATM function, this variant showed a functionally normal result (Lee KS et al. Cell. 2025 Sep;188:5081-5099.e27).
