NM_021930.6(RINT1):c.554C>T (p.Pro185Leu) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P185L variant (also known as c.554C>T), located in coding exon 5 of the RINT1 gene, results from a C to T substitution at nucleotide position 554. The proline at codon 185 is replaced by leucine, an amino acid with similar properties. In one study, this alteration was observed in 0/2024 breast and/or ovarian cancer cases and 1/1886 controls (Li N et al. Breast Cancer Res Treat, 2016 09;159:385-92). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
