Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001134363.3(RBM20):c.3267dup (p.Ile1090fs), citing Ambry Variant Classification Scheme 2023: The c.3267dupC variant, located in coding exon 11 of the RBM20 gene, results from a duplication of C at nucleotide position 3267, causing a translational frameshift with a predicted alternate stop codon (p.I1090Hfs*4). This alteration is expected to result in protein truncation or nonsense-mediated mRNA decay. However, loss of function of RBM20 has not been established as a mechanism of disease. Based on the available evidence, the clinical significance of this alteration remains unclear.
