Uncertain significance for Bethlem myopathy 1A — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004369.4(COL6A3):c.8290T>G (p.Cys2764Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine with glycine at codon 2764 of the COL6A3 protein (p.Cys2764Gly). The cysteine residue is highly conserved and there is a large physicochemical difference between cysteine and glycine. This variant is present in population databases (rs779036168, ExAC 0.009%). This variant has not been reported in the literature in individuals with COL6A3-related disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
